دراسة نمذجة جزيئية لتأثير مجموعة من المركبات المستحصلة لبعض الزيوت العطرية على إنزيم KAS I في جراثيم السّل

الملخص

الهدف: إجراء دراسة نمذجة جزيئية على مجموعة من المركبات المستحصلة من الزيوت العطرية من أجل اكتشاف مركبات طبيعية مضادة لعصيات السل.

المواد  والطرائق: تمت عملية النمذجة الجزيئية بواسطة برنامج iGemdock باستخدام المعيار Standard Docking . حيث تطلبت تحميل بنية الإنزيم الهدف  KAS I من موقع Protein Data Bank وثم تم جمع صيغ المركبات الكيميائية ل 89 مركب مستخلص من الزيوت العطرية  من موقع Pubchem  وتم رسم الصيغ ببرنامج ChemDraw ومن ثم تمت عملية الارساء Docking بين الإنزيم الهدف KAS I والمركبات المدروسة، وبالإضافة إلى ذلك تم تطبيق قواعد لايبنسكي وفيبر على مجموعة المركبات المدروسة من أجل التنبؤ بالتوافر الحيوي لهذه المركبات. حيث تم جمع بيانات هاتين القاعدتين من موقع Pubchem .

النتائج: أظهرت النتائج أن 19 مركب حقق طاقة ارتباط أعلى من طاقة ارتباط المركب العياري وتشير هذه النتائج إلى أنه من الممكن اقتراحها كمركبات مضادة لجراثيم السل.

الخلاصة: إن هذه الدراسة النظرية ألقت الضوء على بعض المركبات الطبيعية منها Apiginin، Stearic acid، Oleic Acid، Arachidic Acid الموجودة في بعض الزيوت العطرية والتي كانت ذات طاقات ارتباط مرتفعة مقارنة مع العياري حيث بلغت طاقته 78.1 KJ/mol -، وبلغت طاقات ارتباط المركبات على التوالي(-104.81 90.23­-،-89.62،-89.38) وبالتالي يمكن اعتبارها ركائز جديدة في استهداف العصيات والحد من مقاومتها العالية للصادات الحيوية المستخدمة.
