دراسة نمذجة جزيئية للتأثير المثبط لمجموعة من القلويدات على أنزيم AchE لدى الإنسان لعلاج الزهايمر

الهدف: إجراء دراسة نمذجة جزيئية على مجموعة من القلويدات التي لها تأثير مثبط لإنزيم أستيل كولين إستراز AchE في دماغ مريض الزهايمر.
المواد والطرائق: تمت عملية النمذجة الجزيئية بواسطة برنامج iGemdock بإستخدام المعيارينstandard وaccurate docking.
حيث تمّ تحميل بنية الأنزيم الهدف AchE المتواجد في الدماغ لدى مرضى الزهايمر من موقع Protein Data Bank وثم تمّ جمع المركبات الكيميائية لـ 101 مركب قلويدي من موقع Pubchem وتمّ رسم الصيغ ببرنامج ChemDraw ومن ثم تمّت عملية الإرساء Docking بين الأنزيم الهدفAchE والمركبات المدروسة، بالإضافة إلى ذلك تم تطبيق قواعد لابينسكي وفيبر على مجموعة المركبات المدروسة من أجل التنبؤ بالتوافر الحيوي لهذه المركبات. حيث تمّ جمع بيانات هاتين القاعدتين من موقع Pubchem.
النتائج: أظهرت النتائج أن 20 مركب حقق طاقة ارتباط أعلى من طاقة ارتباط المركب العياري وهو هوبرزين A وتشير هذه النتائج إلى أنه من الممكن اقتراحها كمركبات مثبطة لـ AchE في خطط علاج مرضى الزهايمر.
الخلاصة: إن هذه الدراسة النظرية ألقت الضوء على بعض القلويدات ودرست دورها كركائز جديدة في استهداف أنزيم AchE والحد من ترقي أعراض مرض الزهايمر
